TYPHOID FEVER DIAGNOSTIC
Assay Procedure
Typhoid fever is an infectious disease caused by bacterium, Salmonella typhi.
It continues to be a major health problem especially in the Asia Pacific region,
the Indian subcontinent, Central Asia, Africa and South America. Definitive
clinical diagnosis of typhoid is unreliable because typhoid fever symptoms
mimic other diseases with fever that are common in this part of the world.
Clinical presentation very tremendously among patients and cover a wide
spectrume, hence the need for a good laboratory test.
Step 1
Add patient serum sample
to pre-dotted antigen strip.
Wash away unbound
patient antibody.
Step 2
Add HRP conjugated
secondry antibody. Wash
away unbond secondry
antibody.
Current methods to diagnose typhoid are by isolation of S. typhi from body
fluids and by detection of antibody using the Widal test. Both methods are
time-consuming with variable sensitivity. The culture method has a sensitivity
of 48-78% but among the pediatric age group the sensitivity could be lower.
Although the Widal test is widely used, its many limitations lead to difficulties in
its interpretation especially in areas of high endemicity.
Step 3
Add substatre and read
results.
We produce two types of diagnostics test kits for typhoid fever - Typhidot and
Typhidot Rapid test kits. Typhidot is using a more sensitive Enzyme
Immunoblot Assay; while Typhidot Rapid test kits are using a faster
Immunochromatography Assay.
Interpretation Of Result :
Typhidot
Typhidot is a revolutionary methos of diagnosis. Invented in Malaysia, it is the
first known qualitative antibody detection test design for rapid diagnosis of
typhoid fever. Using the Dot EIA technique, presence of IgM & IgG antibodies
against a specific antigen on the outer membrane of S. typhi are detected.
Typhidot has been evaluated by many endemic areas including Malaysia,
India, Indonesia, Pakistan and Philippines. A control clinical field trial among
febrile children admitted to a hospital in an endemic area showed that Typhidot
when compared to culture is sensitive (95%) and did not show any
cross-reactions.

Step 1
Add serum/ plasma/
whole blood to square
well. For whole blood this
is followed by addition of
one drop buffer
Step 2
Up to area A, add 3 drops
buffer to oval well.
The test kits are designed for the qualitative detection and differentiation of
specific IgM or IgG/ IgM antibodies against specific S. typhi antigen.
Step 3
Pull clear plastic tab and
add one drop buffer to
square well
