Feature stainless steel construction for long life and corrosion protection.
Multiple cycle selections and microprocessor control ensure that each load is processed
efficiently and with the minimum energy required to meet cleaning requirements.
Operation is simple and loading is easy and flexible. A delayed-start feature allows the
washers to operate automatically unattended during off-peak hours.
Uses a maximum of 15L (3.9gal.) of tap water. Low water requirement and precise
detergent and neutralizer dosing minimize chemical costs
Spindle-ready models use an existing deionized water supply and can accommodate
either one or two direct injection spindle racks to clean up to 44 flasks.

Changing Cycle Parameters:
1. Press Cycle to be changed, then press
MENU key, LED will light.
2. To change CycleType, press Enter,
then up arrow to see all types, press
Enter again to select a type.
3. To change other parameters on this
Cycle, use up arrow to scroll to the
next cycle parameter to be modified.
4. To change that value, Press Enter key,
“asterisk” key then lights up.
5. Use up/down arrows to change the
value (must be within allowable range
see pg 14)
6. Press ENTER key when done, then
continue up arrow to next parameter
7. Press ADV key to exit and return to
the idle screen for normal operations.

STARTING A CYCLE, be patient
1. Press the keypad for the desired cycle, the
LED for that pad will turn on. For cycles
above 4, press the up arrow and keypad
numbers 1-4 (cycles 5-8)

confirmation with the Door Closed LED.

Wait ..…..The “START” button be not be
operative until the jacket temperature is
within 6 F of the cycle operating
temperature.

To view the Jacket temperature, press the
Upper left hand heart on the Primus
symbol.
5.
When the jacket temperature has been
reached, the READY and START LEDs will
be lit.
6.
Press the START button and hold for a few
seconds to begin the cycle. The READY
and START LED indicators will go out,
other cycle indicator LEDs will follow the
Cycle as it runs.

Autoclave: Safety Guidelines, Primus, BRDG
1. Never overload the Sterilizer, sterilization may never be achieved.
2. LIQUID cycles are only for the sterilization of Liquids, DO NOT USE
ANY OTHER CYCLE for Liquids.
3. When processing LIQUIDS, use containers with vented closures..do not use
ordinary glass bottles or any containers not designed for this purpose.
4. NEVER sterilize sealed containers in a cycle other than a LIQUIDS
cycle, rapid pressure changes in other cycles can cause explosive results.
5. NEVER obstruct the drains in the bottom of the chamber, always load in a
manner where no objects will touch or move to chamber walls.
6. ALWAYS use “hot gloves” when loading or unloading the sterilizer,
before or after a cycle…. ALWAYS assume the door, walls, racks,
and/or gurney are HOT!
7. ALWAYS place “hot gloves” on the ends of a loading cart
to indicate to others that the cart and labware are HOT!

WEEKLY – Execute this BLOW OUT protocol to
maintain boiler when unit is at operating
condition
1. Turn OFF boiler (toggle switch on rear of boiler)
2. Turn OFF mail 208V power switch (left front wall)
3. Turn OFF autoclave (control panel)
4. Turn OFF Main water supply valve at rear wall
5. Unplug all three 110V power cords at rear
6. Remove Right side Autoclave Stainless Panel
7. Using a protective glove locate Blow-Out Valve
(Green) at lower rear
8. Open valve slowly about 30 degrees
counterclockwise
9. Observe water/steam exiting to drain on wall
behind Millipore…. Wait until steam subsides
10. Open valve another 30 degrees, wait, and repeat
again until valve is fully open (aligned with the
exit pipe)
11. Return Valve to closed (pointing up) position
12. Turn ON main water supply valve
13. Turn ON main 208V switch and boiler toggle
switch (left rear)
14. Re-insert 110V power plugs
15. Watch Glass tube on side of boiler to ensure it
has refilled to acceptable level

Recommended Method (BobM)
1. Place empty spool in the area of the
hubs
2. Use a screwdriver to open/expand
the width of the open slot to make it
easier to snap over the hub ends
3. When empty spool is in place, fold
paper into a “V”, insert into spool
slot
4. Press FEED to take up some paper
on spool.

Autoclave : Tuttnauer Basic Operating Instructions
1.
Open the main steam inlet valve and watch the jacket pressure gauge on the front
panel. When the jacket pressure reaches 2.3 bars, the sterilization cycle can be
started. (condensate may exit through the jacket steam trap during this period)
Open the door by turning the control wheel handle counter clockwise to release the
locking arms.
Load the material to be sterilized per the guidelines in this protocol
Close the door by pushing towards the frame and turning the wheel handle
clockwise until the “Door Closed” lamp is lit.
On the control panel, chose the sterilization program according to the materials
loaded. The light for the program should be on with parameters displayed.
Press the START key to begin the cycle. Lights on the bottom indicate the stage of
the cycles in progress.

Air is purged from the chamber at the beginning of the sterilization cycle.
Steam under pressure is introduced into the chamber and the temperature rises to the desired level.
The autoclave is maintained under pressure at a high temperature for a predetermined time.
Steam is then exhausted from the chamber at the end of the cycle.
A buzzer will sound and the START light will turn off at the end of the cycle. The
printer will print a summary of the cycle.
Open the door by turning the wheel handle counter clockwise.

• Safety valve – opens as back-up protection to reduce chamber pressure in the
event of exceeding 262 kPa

Vent Valve – needed if chamber pressure should exceed 241kPa, the vent
valve will open and the P-2 alarm will display.

from the Autoclave

Overheat Protection – chamber temperature is set so as not to exceed 149 C and has
additional overheat protection if temperature reaches 180 C.
2. Electrical power interruption – in case of an electrical power failure during the
sterilization cycle, pressure in the chamber is automatically vented to the atmosphere.

Autoclave: Sterilizing Media in Flask ; Removing and Cooling
7a. When the autoclave is finished and the inside
pressure has dissipated, remove your flask of media
using orange hot gloves.
7b. Swirl the media gently because the agar may
settle to the bottom of the flask during autoclaving.
8a. Place the media into a water bath set at 55-60C,
which is the temperature it should cool to for pouring
the plates. (Note: If the solution is not placed in a water
bath, cool until you can just hold your bare hands on
the bottom of the flask.
8b. If it begins to solidify, it will have to be re-melted by
autoclaving again for about 5 minutes.

Sterilization – the highest level of disinfection; the killing of all living

Testing whether sterilization has occurred during autoclaving is done
using spores (dormant microorganisms)
– Spore strips are dried pieces of paper to which large number of
pathogenic , heat-resistant Bacillus thermopholus spores are
adhered.
– After autoclaving the strips are transferred to growth medium, if
successful autoclaving, then the spores will not grow.

Autoclaves: Cycles, Gravity vs. Vacuum
Gravity Cycles: Displacement Autoclaves – The Old Workhorse
Traditional models rely upon gravity to displace the air from the sterilizing chamber. Steam is much lighter than air and, as it is
injected into the pressure chamber, it forces air downwards, trapping it in a sink. A system of valves and taps bleed out this excess
air, allowing the steam to sterilize the contents of the chamber.
These autoclaves are the cheapest option and they are very easy to use. As a result, they are common in kitchens and manicurists,
where they are more than adequate. However, for sterilizing porous or wrapped materials, displacement autoclaves are not efficient
enough. Hot air may be trapped inside, allowing pockets of microbes and other pathogens to evade the sterilization cycle.
Vacuum Autoclaves – The Safer Solution to Sterilization
For complex sterilization, or in areas where complete sterility is necessary, a vacuum autoclave is the superior choice. Vacuum
autoclaves evacuate the air, with a vacuum, and steam is then injected into the chamber. This allows the steam to penetrate porous
materials, wiping out any pockets of pathogens in hard-to-reach areas.
After this steam sterilization stage, the pump again evacuates the chamber, drying the autoclave contents thoroughly. This added
stage prevents germs and prions being retransmitted by trapped moisture. Vacuum autoclaves are the preferred option in medical
institutions, as a more effective alternative to the simpler displacement autoclaves.
Many hospitals use disposable medical equipment, to prevent the transmission of CJD variants, but the contaminated waste is still
autoclaved before landfill. Autoclaving is a more environmentally friendly method than traditional incineration, which ejects toxic
gases into the atmosphere.

The Amsco Lab 250 sterilizer was designed with the needs and
the budget of the laboratory research market in mind. The lab
series sterilizer provides the same time-proven, safe, reliable
chamber and stand assembly as the Century® sterilizer that has
been offered to the healthcare and life sciences markets since
1992.
The Amsco Lab 250 features an Allen-Bradley MicroLogix"
control and a full-color operator interface. The unit has a
chamber size of 20 x 20 x 38 inches (508 x 508 x 965 mm) and
is equipped with gravity and liquid type cycles as standard.
Optional prevacuum and isothermal cycles are also available if
required. A manually operated vertical sliding door is combined
with the non-lubricated door seal for reliability and ease of use.
The drain line piping package meets or exceeds the maximum
water temperature requirements of the industry - 60ºC (140ºF).

Allow 6-10 minutes for the Bacti-Cinerator to reach an
optimum sterilizing temperature of 815°C (1500°F) throughout the interior of the core.
5-7 seconds of exposure sterilizes loops, needles and culture tubes. Unit is not intended
for use with scalpels, forceps or sharp instruments.
Use only insulated loop holders when inserting loops into the heater element.
Suitable for anaerobic procedures in chambers and hoods.
Yellow Light indicates that the heating element is ON. Note, the Core element may
remain very hot for 30 minutes after it is turned off. Allow 1 hour of cooling before
cleaning or storage.
Avoid any contact with the heat shield or end caps while the Sterilizer is on.
Do not immerse the sterilizer in water or other cleaning solutions.
Periodically inspect the element for cracks (yellow-orange fissures) and wear.
